Ion Exchange Columns Ion-exchange chromatography columns separate and identify compounds based on their ionic or electrostatic interactions. These columns are widely used for the purification of proteins and peptides, as well as nucleic acids. Anion exchange columns will exchange negatively charged molecules while cation exchange columns will exchange positively charged molecules. In addition, chromatography resins can be further categorized as weak or strong ion exchangers, a distinction determined by the effects of buffer pH. The interaction of target molecules with the stationary phase media varies according to overall charge, charge density, and surface charge distribution.
