Protein Labeling Reagents

Protein labeling reagents are available from a number of suppliers for the attachment of conjugates, dyes, and other moieties to proteins. Among these reagents are compounds with reactive groups that facilitate covalent binding with proteins. NHS (N-Hydroxysuccinimide) esters, for example, will attach to amino groups in proteins, such as lysine residues. Maleimide reactive groups selectively react with protein residues containing a sulfhydryl group, such as cysteines. Conjugates with hydrazide reactive groups attach via carbonyl groups. These labeling reagents, which usually proceed with simple reaction and purification steps, are useful in attaching conjugates such as gold, biotin, fluorophores, and dyes to purified proteins and antibodies.
