Principle of the Assay::The Human CD321 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD321 in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Human CD321 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD321 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD321 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD321 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Junctional adhesion molecule A (JAM-A) is a protein that in humans is encoded by the F11R gene. It is mapped to 1q23.3. This gene is an immunoglobulin-like molecule that colocalizes with tight junctions in endothelium and epithelium and is also found on blood leukocytes and platelets. JAM-A plays an important role in the regulation of tight junction assembly in epithelia. In addition, it can act as a receptor for reovirus, a ligand for the integrin LFA1, involved in leukocyte transmigration, and a platelet receptor. JAM-A has a nonredundant role in controlling DC motility, trafficking to lymph nodes, and activation of specific immunity
