The proteasome is the main protein degradation machinery in cells. To study the effect of proteasome inhibition on the cellular proteome, I used an inhibitor that blocks the chymotrypsin-like activity of the proteasome. Further, using this luminescence-based assay kit, I confirmed the decrease in proteasome activity.

The assay is luminescence based and so requires specific plates and reader. Store the prepared assay solution in aliquots for longer stability.

Results Summary

Cells were plated in 96-well culture plate. After 8 hr of inhibitor treatment, pre-mixed assay buffer was added in equal volume to the sample and incubated for 10 min at RT. The supernatant was transferred to white opaque microplate and luminescence recorded using plate reader.
