In Vivo Transfection

Transfection Reagents Transfection reagents are used to introduce DNA or RNA into eukaryotic cells. Transfection reagents help overcome the barrier caused by the negative charges of both the nucleic acids and the cellular membrane by giving an overall positive charge to the DNA or RNA through the formation of transfection complexes. This interaction is necessary to trigger cellular uptake of the complexes and subsequent release from vesicles into the cytoplasm. Transfection reagents are often cationic lipids or polymers and include calcium phosphate and diethylaminoethyl (DEAE)-dextran. Visit the supplier pages for more product information.

As with most cases regarding transition from in vitro to in vivo, In Vivo transfection is technically complicated. Some factors to take into account include DNA/RNA stability, efficiency of delivery into the body, and tissue/organ specificity. Fortunately, commercial manufacturers have designed kits and reagents especially for DNA or RNA delivery into the live animal. Lipid and non-lipid formulations are available, and many feature low toxicity and high delivery efficiency. Supporting data and publication references are also provided.

  • FemtoJet 4x

The goal of our research is to create a more efficient metastatic prostate cancer GEMM and we will be using both the femtojet and transferman to inject virus cells into our current mouse model and have the cancer cells become more precise. In the past, you could not differentiate the cancer cells, and with this new method of injection, we are able to create single colonies of cancer cells. This technique of using microinjection is also helpful for work with gene targeting and IVF. The goal of this product is to ensure that virus reaches the tissue and produces metastatic disease. By using the product, instead of by hand, creates more reproducible spread of cancer. This product was chosen because they could help us obtain the results we wanted and were most cost effective.
