We used the Lactate-Glo Assay Kit to quantify lactate concentrations in cultured primary astrocytes, mouse prefrontal cortex, cerebrospinal fluid, and serum while investigating astrocyte-specific regulation of brain energy metabolism following Npas3 deletion. The assay generated reproducible luminescence signals across diverse biological samples and enabled detection of physiologically relevant metabolic changes.
Luminescent lactate assay
Primary mouse astrocytes, mouse prefrontal cortex, cerebrospinal fluid, and serum
Protein normalization of tissue and cell lysates improved comparison between experimental groups.
The assay showed excellent sensitivity and reproducibility across cultured cells and multiple tissue-derived samples. The luminescent signal was stable, easy to quantify, and enabled reliable detection of lactate alterations associated with astrocyte-specific Npas3 deletion. The kit performed consistently with low background and required minimal optimization.
