A mouse model with I4895T mutation in type 1 ryanodine receptor displays myopathy including muscle weakness and atrophy. In vivo protein synthesis with puromycin labeling showed decrease of newly synthesized proteins compared to WT.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Skeletal muscle

Primary Incubation

1:1000, 2hs at R.T (or overnight at cold room)

Blocking Agent

Bio-Rad EveryBlot Blocking buffer

Secondary Incubation

1:10,000, 1 hour at room temperature (StarBright 700)

Tertiary Incubation

No tertiary incubation

Detection

Bio-Rad ChemiDoc MP Imaging System

Results Summary

This is the assay for the detection of newly synthesized proteins (SUnSET). IP injection of puromycin into mice for 35 min labels newly synthesized proteins with puromycin. Puromycin Ab detects these proteins.
