My general research aim is to investigate the role of autophagy in neurodegenerative and infectious diseases, using stem cells and stem cell-derived human cellular platforms. The product is a kit for evaluating the percentage of dead and alive cells in a population via fluorescence imaging.

If fluorescent microscope is available, make sure the reagents already worked.

Results Summary

We imaged the cells after staining them with both reagents from the kit. The incubation time that was suggested by the manufacturer was not enough to stain the cells with Hoechst and therefore we needed to prolong the incubation to 30 minutes.

Summary

The Good

Very easy-to-use kit with just a drop or two needing to be added to the cells.

The Bad

15 minutes might not be enough to see good staining

The Bottom Line

If you don't have a fluorescent microscope in the lab, it might be a good idea to prolong the incubation (asked the manufacturer and they confirmed that incubation is in RT) up to 30 mins at least in 1 sample. In our lab, even after 30 mins some cells were not stained properly though.
