Total RNA
5 microgram of good quality total RNA was treated with DNAse to degrade the DNA in the sample. Now the RNA is used for cDNA synthesis. The RNA is incubated at 37 degrees with 2XRQ buffer and mRQ enzyme to the final volume of 10 microliter. After one hour the sample is diluted with 90 microliters of nuclease free water.
Always use a good quality total RNA
The miRNAs can be quantified using miRNA specific forward primer and the 3' universal primer provided in the kit. The U6 primers provided in the kit is used as the reference gene.
