BD's LSR-II analyzer has the capacity to support up to 7 lasers and has 56 positional choices for selection of the detectors; however, the model I am reviewing has 3 lasers and 9 detectors. It is now ~ 8 years old, and it shows! One laser is nearly dead, and the machine needs regular maintenance.Yet, it is a reliable instrument most of the times and provides digital output (FCS 3.0 and 3.1 files). The software (Diva 6.0) is OK, but not the easiest to use. I think BD can learn a thing or two from FlowJo.

The machine is quite robust, even when the sample is poor. Priming and cleaning with 10% bleach usually unblocks the flow.

Tips

You need to understand the basics of flow cytometry, otherwise you are wasting your time.

Results Summary

Results are very much dependent on your assay. As I always say: "garbage in, garbage out". Make sure you prepare the samples properly including single-color compensation controls. Always talk to someone who has experience with the machine and the technique before you even run your assay!
