Fig 1: NG and nGO decrease the synthesis of NF-?B related proteins. (A) Antibody array analysis of proangiogenic cytokine synthesis in U87 and U118 glioma cells with or without treatment (C; control) with graphite nanoparticles (NG) and graphene oxide nanoplatelets (nGO). The full array map and uncropped images are available in the supplement. Nanoparticle treatment results in decreased synthesis of GROa, IL-6, IL-8, and MCP-1 in U87 but not U118 cells. Transcription factor assay analysis of the activity of p50 and p65 NF-?B subunits in the nuclear fraction of U87 (B,C) and U118 (F,G) cells. Nanoparticle treatment of U87 cells decreased the activation of the p65 subunit and increased the activation of the p50 subunit. Analysis of STAT3 activation showed decreased STAT3 phosphorylation after nanoparticle treatment of U87 cells (D) but increased phosphorylation after treatment of U118 cells (H). The total STAT3 protein level was not changed (E,I). Statistical significance is indicated with different superscripts (one-way ANOVA; P < 0.05). RU, relative units.
