Immunofluorescence of Presenilin1 in Human Brain cells with Presenilin 1 Polyclonal Antibody (Product # PA5-20376) at 20 µg/mL.

Western blot was performed using Anti-Presenilin 1 Polyclonal Antibody (Product # PA5-20376) and a 53 kDa band corresponding to Presenilin 1 and ~20 kDa band corresponding to C-terminal fragment of Presenilin 1 was observed across cell lines tested. Membrane enriched extracts (30 µg lysate) of T-47D (Lane 1), MCF7 (Lane 2), HT-29 (Lane 3), SH-SY5Y (Lane 4) and Daudi (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (2 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
