Western blot was performed using Anti-Furin Polyclonal Antibody (Product # PA1-062) and a 87 kDa band corresponding to Furin was observed across all cell lines and tissues tested except Daudi and Ramos which are known to be low expressing cell lines (DOI: 10.1099/0022-1317-75-10-2821). Whole cell extracts (30 µg lysate) of HEK-293 (Lane 1), HaCaT (Lane 2), BeWo (Lane 3), SK-O-V3 (Lane 4), Daudi (Lane 5), Ramos (Lane 6), Mouse Liver (Lane 7) and Rat Liver (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Expression of Furin was observed in HEK-293, HaCaT, BeWo, SK-O-V3, Mouse Liver and Rat Liver but not in Daudi and Ramos which are known to be low expressing cell lines (DOI: 10.1099/0022-1317-75-10-2821).
