Assay Principle
Alpha-galactosidase (encoded by GLA, also called melibiase) is a lysosomal glycoside hydrolase that removes terminal alpha-galactosyl residues from glycolipids and glycoproteins. An alpha-galactosidase assay typically relies on a synthetic chromogenic or fluorogenic substrate: in the colorimetric version, the enzyme hydrolyzes a p-nitrophenyl-alpha-D-galactopyranoside substrate to release p-nitrophenol, a yellow product with absorbance typically read around 400 to 405 nm on a microplate reader. Fluorometric versions use an analogous fluorogenic glycoside substrate and are read on a fluorescence plate reader, offering higher sensitivity for samples with low enzyme activity. Both formats compare sample signal to a standard curve, usually built from a p-nitrophenol or equivalent fluorophore standard, to convert absorbance or fluorescence into enzyme activity units.
