Assay Principle
Acetyl-CoA carboxylase (ACC) is a biotin-dependent enzyme that catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting and committed step in fatty acid synthesis. Most commercial ACC assay kits use a colorimetric phosphate-release format: ACC reacts with acetyl-CoA, bicarbonate, and ATP to generate malonyl-CoA, ADP, and inorganic phosphate, and the amount of phosphate released is directly proportional to enzyme activity. A dye reagent forms a colored complex with the liberated phosphate, and absorbance is read on a microplate reader, typically somewhere in the 630 to 670 nm range depending on the specific dye chemistry used. A standard curve of known phosphate or activity units is run alongside samples to convert absorbance into an activity value.

Since the assay depends on an oxidation-sensitive dye, kits that specify shorter working-reagent windows or single-use aliquoting may be more forgiving for labs running batches over multiple days. It is also worth confirming whether a kit's detection range and sensitivity match the expected ACC activity in your sample type, since reported ranges can vary by an order of magnitude between suppliers. As with any activity assay, it's good practice to review the full protocol before deciding on a kit, since the manual often reveals practical details, such as incubation times, control requirements, and data analysis steps, that aren't captured in the product summary alone.
