Transfer Buffers Transfer buffers are necessary reagents for blotting techniques in order to facilitate the binding of the protein to the membrane, as well as for the elution of proteins from a matrix. In Western blotting, the transfer buffer is used to fully immerse the gel and membrane sandwich prior to the wet method of electrotransfer. Transfer buffers, such as Tris-Glycine, generally resemble the buffering composition of gels and running buffers. However, commercial transfer buffers feature optimized formulations that are designed to enhance the retention of proteins, such as those with low abundance. Tris-CAPS transfer buffers are commonly used for semi-dry blotting systems, while Towbin and CAPS buffers are used for protein sequencing, glycoproteins, and small proteins and peptides.
