
TIRFM is a well-established technique for live cell imaging that makes use of total internal reflection (TIR) to produce extremely high contrast, low background images. The electromagnetic field of the light extends into the sample beyond the interface by some one to two hundred nanometers yielding a very thin section of the specimen to undergo fluorescence excitation.
