AteloGene™ Systemic Use

AteloGene™ Kit for Systemic siRNA Delivery in Live Animals. Important Note: This kit (product KOU-1393) is for systemic (intravenous) injection only. The atelocollagen in this kit will NOT be gelated in vivo, insuring efficient siRNA delivery throught the cicrulatory system. For localized siRNA delivery, please see AteloGene™ Kit for Localized siRNA Delivery Into Live Animals (product KOU-1392). BACKGROUND As a cationic polymer, atelocollagen is a proven, effective carrier for the delivery and cellular uptake of siRNA in vivo as shown by specific inhibition of gene expression or biological processes (1). As a world leader in collagen technology, KOKEN Co., Ltd (Tokyo, Japan) has developed the AteloGene™ system for atelocollagen-based siRNA delivery in vivo. (1) Cellular delivery in vivo of siRNA-based therapeutics., Aigner A., Curr Pharm Des. 2008;14(34):3603-19. PMID: 19075737 AteloGene™ KIT FEATURES * Specialized formulations for localized (subcutaneous) or systemic (intravenous) injection. * Simple, convenient, fast preparation. Mix and inject. * Efficient in vivo transfection * Persistant resistance to RNase * Non-toxic. High biocompatibility. KIT CONTENTS AteloGene prefilled syringe (600 µL × 2) 10× siRNA buffer (3 ml x 1) sterile water (3 ml x 1) 2 ml microtube for mixing (x2) 1 ml syringe for injection (x2) 18G needle for mixing and filling (x4) 26G needle for injetion (x2) Instruction manual Sufficient material for 10 mice PROTOCOL OVERVIEW Eject AteloGene material from prefilled syringe into mixing tube. Add siRNA solution. Mix. Centrifuge. Withdraw mixture into injection syringe. Inject at administration site (KOU-1392) or into tail vein (KOU-1393). NOTE: This product is for non-human research use only. Application to humans is strictly prohibited. What is Atelocollagen? Atelocollagen from the Koken Co. is prepared from highly purified collagen extracted from BSE-free calf dermis and further processed by protease treatment to remove short stretches of amino acids present at both the N- and C-termini of collagen strands (telopeptides). As telopeptide amino acid sequence diversity confers most of collagen's immunogenicity, atelocolagen displays low immunogenicty and other desireable features including high biocompatibility, and biodegradability, and high plasticity, yet retains many physical properties of native collagen. Protease treatment also digests protein contaminants resulting in a highly purified material that has found wide application in medicine, medical devices, cosmetics, and cell culture. Koken, a world leader in collagen technology, has used their expertise to produce an outstanding array of collagen-derived products for use in cell culture (AteloCell products) and as an exciting new vector for the effective in vivo delievery of siRNAi(AteloGene Kits).
