Description

The X-CLARITY™ Polymerization System is a standalone, automated system developed to simplify hydrogel-tissue hybridization, a crucial step for optimal tissue clearing with the CLARITY method. To initiate polymerization, a fixed tissue sample saturated with a hydrogel monomer and thermal initiator solution is heated. The rise in temperature causes the thermal initiator to release free radicals and initiate radical polymerization of the hydrogel monomers, covalently linking the biomolecules in the tissue sample to a sturdy hydrogel network, preserving molecular information and structural integrity. The process is done in a strictly anaerobic environment as oxygen is a strong inhibitor of polymerization. Manual hydrogel polymerization requires multiple steps, which include degassing the sample in a desiccator chamber with a vacuum pump, replacing the oxygen with an inert gas or mineral oil, and then heating the sample in a water bath. The X-CLARITY™ Polymerization System combines these steps into one. Multiple samples can be placed in well plates or conical tubes for rapid, efficient, and high-throughput sample processing. Users can control polymerization by adjusting vacuum strength, temperature, and a timer through a simple touchscreen interface.
