  • Western blot with HMGB1 Antibody in CHO Cells

    This antibody was used to monitor equal protein loading in all wells (house keeping gene control). Methodology: CHO cell lysates were prepared in RIPA lysis buffer. 50 µg of clarified lysate was loaded on a 10% Tris-Glycine gel and SDS-PAGE was performed. Proteins were transferred onto a nitrocellulose membrane using semi-dry transfer method. Ponceau staining was done to visualize proper protein transfer. The membrane was blocked in blocking buffer (Odyssey Blocking Buffer) for 30 min at room temperature. HMGB1 antibody was diluted (1:1000 dilution) in blocking buffer and incubated with the membrane for 1 hr at room temperature. Membrane was washed thrice in wash buffer (1xTBS-T (0.1%)) and then incubated with secondary antibody – LI-COR IRDye 800 (1:5000 dilution) for 1 hr at room temperature. Membrane was washed thrice in wash buffer and the signal was captured on a LI-COR Odyssey imager. Rating: Very good antibody with low signal/noise ratio
