CD33 is typically positive on monocytes (bright) and neutrophils (intermediate/dim) but completely negative on lymphocytes. We extensively use this antibody as one of the markers for acute myeloid leukemia specimens.

Results Summary

Although PerCP-Cy™5.5 is a known "dim" fluorochrome, we were expecting better results from this antibody than what we got. The resolution of the three populations (monocytes, granulocytes, and lymphocytes) is just about enough to separate them. This is even weaker when multi-color assays are designed with this antibody.

Summary

The Good

Stains cells appropriately.

The Bad

Dim expression on positive cells.

The Bottom Line

PerCP-Cy™5.5 should be reserved for markers that are highly expressed on the cells. Using it with CD33 probably does not work as well as needed.
