COX is the terminal enzyme of the respiratory chain that, catalyzes the transfer of electrons from reduced cytochrome c to molecular oxygen. We deleted the Cox10 gene that encodes for one of the accessory proteins of Complex IV. We found that astrocyte-specific deletion of Cox10 and resultant deficiency of Complex IV and oxidative phosphorylation attenuated novel object recognition and trace fear conditioning. The findings suggest that astrocyte oxidative phosphorylation may contribute to behaviors associated with greater cognitive load and stressful conditions.

Experimental Design and Results Summary

Application

BaseScope ISH

Starting Material

Mouse brain sample

Tips

Keep in mind that dilution should be 1:50

Results Summary

To assess regional changes in expression of Cox10 mRNA, we used BaseScope in situ hybridization and found an approximately 50%-decrease in expression of Cox10 in the dentate gyrus of the hippocampus, the CA1-CA2 areas of the hippocampus and the caudate putamen.
