My goal was to understand interactions between cells and nanoparticles, and the biological effect of those nanoparticles on cancer and non-cancer cells in vitro. For that, I've used a lot of fluorescence and brightfield microscopy in assays such as scratch assays, proliferation rate, cell viability, and immunofluorescence, all acquired with this equipment.

Experimental Design and Results Summary

Application

Brightfield microscopy for scratch assay/wound healing; Fluorescence microscopy for EdU proliferation assay, staining of many proteins using antibodies; Absorbance of light for cell viability assays such as MTT and calcein; DNA quantification after plasmid extraction; Cell lineage health checks such as mycoplasma test using Hoechst; Whole-tissue acquisition of mouse brain sections.

Understand how the autofocus of the equipment work makes it easier to acquire data.

Summary

The Good

Very robust, never broke. Has a good balance between quality of the data and speed of acquisition; and it performs many tasks, because with only one equipment, you can acquire many types of data (image, absorbance, luminescence, UV-vis).

The Bad

Take some time to understand how it works; You have to manually change the optical distance in the objective lenses.

The Bottom Line

Robust and multitasking, acquired all my master's and Ph.D. data using it.
