Both human iPSC and human ESC were cultured in feeder-free conditions on 1:100 matrigel-coated culture plates. ReLeSRTM reagent was successfully used to passage stem cells in small clumps (5-30 cells) to new culture plates for over 10 passages.

Leave ReLeSRTMon cells for 4-5min max, avoid over pipetting to maintain small clumps of cells.

Results Summary

Cells incubated with ReLeSRTM for 5 min before neutralization with culture media was lifted easily off culture plates with minimal pipetting. Clumps of 5-30 cells remained for passage to new plates, and cells quickly formed colonies with round morphology thereafter.

Summary

The Good

Fast and simple, does not require centrifugation for passage, and allows passage of stem cells in small clumps preventing single cell dissociation.

The Bad

More expensive than alternatives like TryplE, less preferable for single cell dissociation and counting.

The Bottom Line

Great reagent for fast and effective stem cell passage in small clumps at a decent price. Would highly recommend for long term culture of stem cells.
