The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. The goal of this experiment is to determine if Twin-Strep tagged AXIN1 expressed in Sf9 insect cells can be purified by Strep-Tactin resin.

Manufacturer's instructions were followed in this experiment.
Sf9 cells expressing Twin-Strep tagged AXIN1 were lysed and clarified lysates (1-8 mg) were incubated with 50 µL of pre-equilibrated Strep-Tactin Sepharose resin. The incubation time was 2 h at 4 deg C. Protein complexes were eluted in 2x SDS sample buffer and resolved on a 4-12% Bis-Tris gel. Immunoblotting was performed with an AXIN1 antibody. Good enrichment of bait was observed.
