CD13 is expressed on granulocytes and monocytes (the myelomonocytic lineage). We opted for APC conjugate because it is typically bright. Although there are cheaper alternatives, we got this antibody from Beckman Coulter.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human whole blood (RBC lysed)

Primary Incubation

15 minutes at room temperature

Blocking Agent

1% bovine serum

Secondary Incubation

Detection

Red laser (APC channel)

Results Summary

The results are shown in the figure: Granulocytes (pink) stained well. Monocytes are also positive. However, it was hard to resolve the monocytes from the lymphocytes. The use of side scatter helps. We would've liked it to be brighter, but the staining is acceptable.

Summary

The Good

Easy titration. No issue with compensation.

The Bad

Relatively dim.

The Bottom Line

This antibody should work for most people, but if a strong signal is needed, then maybe try a different antibody/fluorochrome. Otherwise, it works OK.
