We used this kit to analyze the inflammasome activation by measuring the activity of caspase-1 in the culture supernatants of human macrophages. Different from the immunoblot assay that usually needs to concentrate culture supernatants or precipitate proteins from supernatant before the detection of cleaved caspase-1, Caspase-Glo® 1 Inflammasome Assay kit can directly detect the caspase-1 activity with a small volume (50-100 ul) of original samples, this it is easier and more sensitive. We have successfully used this kit in our studies and the tested results have been published.

We treated differentiated human macrophages with or without LPS/ATP for 4 hours, the activity of caspase-1 in the culture supernatants was measured by using Caspase-Glo® 1 Inflammasome Assay kit. As expected, treatment of human macrophages with NLRP3 inflammasome inducer LPS/ATP resulted in increased activity of caspase-I in the culture supernatant as tested by kit. This result was successfully reproduced for many times in our study.

Very sensitive and only need small amount of sample

This kit works great for measuring the caspase_1 activation from a small amount of cell culture supernatants, I highly recommend it.
