The mouse tissues were homogenized, sonicated, and proteins were extracted in RIPA buffer. The extracted proteins were measured, and an equal amount of proteins were resolved in 10% SDS-PAGE. Resolved proteins were further immunoblotted for the detection of SHP2 (phospho Y542) protein using a specific antibody obtained from Abcam. The selection of antibody was based on the merits shown in antibody flyer.
