A high degree of precision is needed for all aspects of this assay (accurate pipetting, plating the same number of cells in each well, etc).

Results Summary

2 different lines of primary human fibroblasts were plated and used for the mito stress test assay. The healthy donor cell line (green) was found to have slightly higher basal respiration as measured by oxygen consumption rate. Both lines had comparable OCR due to non-mitochondrial oxygen consumption+proton leak when oligomycin was used to block ATP synthase. When FCCP is used to disrupt the proton gradient, the healthy line achieved a higher maximum OCR. Both lines had comparable OCR due to non mitochondrial oxygen consumption when Rotenone+antimycin A were used to inhibit complex 1+3.
