I liked this kit as the reconstitution of reagents is fairly simple (need to ensure you are reconstituting the right volumes for the right kit, as they come in different volumes for each kit) and then you can store the 2 ready reagents for a couple of months at -80. Make sure you have precisely 100 uls in each well. As medium tends to evaporate in the incubator, I would usually remove all media before doing the assay and pipette back precisely 100 ul, then proceed to adding the reagent. We also tried to do 2 second readings after adding digitonin. The instructions say make the second reading after digitonin after 15 mins, but we also made another one after 1 hour to ensure the overall cell death number is being optimized. I like the simple data analysis as you just subtract the numbers of the 2 readings. Its slightly harder to run the assay on iPSCs as they grow in colonies and you need to be careful with the seeding density to be similar among the wells. Overall it an easy to use assay though and can be done fairly easily.
