Our lab starting using this machine as our go-to procedure for identifying cells in specific tumor cells ex-vivo. This machine has variety of colors for each antibody so we were able to identify a variety of cell populations and analyze expressions of molecules.

Protocol Overview

We obtain single cell suspension from our samples, followed by surface stain. If we need to stain intracellularly, we would fix/perm our samples before we add the intracellular stain. After we wash samples out with a specific buffer before running on the machine.

Tips

Make sure to clean it well after each use to avoid contamination or clogs!

Features Summary

We had the older generation of this model and this current one is a lot quieter than the previous ones.

The Good

Runs well and quickly. Program that comes with it is also pretty inuitive and easy to export files.

Some clogging and definitely needs maintenance.
