We take frozen sections of mouse whole eyes to examine the different layers of the eye. This vectashield allow us to protect our stained cells on these sections and also adds a DAPI layer.

Protocol Overview

Frozen sections are fixed in 4% paraformaldehyde for 30 minutes and then washed in cold PBS 3 times. Vectashield is dropped sparingly onto the slides and coverslip is added gently.

Tips

Don't use too much or it will squeeze out.

Results Summary

This medium will not fade even over months (if slides are stored in the dark) and clearly stains Dapi in the frozen sections.
