Product Name:

APC-Cy7 Anti-mouse CD4

Catalog Number:

100526

CD4, also known as L3T4 or T4 is expressed on most thymocytes and a subset of T cells, and weakly on macrophages and dendritic cells. It acts as a co-receptor with the TCR during T cell activation and thymic differentiation by binding MHC class II. The clone RM4-5 is a well known anti-mouse CD4 antibody clone and blocks the binding of GK1.4 antibody, which is a known depleting agent of CD4 T cells.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse splenocytes

Primary Incubation

25 minutes on ice

Blocking Agent

CD16/32 antibody for blocking FC receptors

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow cytometry on APC-Cy7 channel

Results Summary

The staining was relevant; however, as usual, the Cy7 modified fluorophores are known to be very bright and bleed significantly into adjacent channels, complicating the compensation parameters. A small level of background shift was also seen in the negative population.

Summary

The Good

Bright and relevant staining

The Bad

Background levels are noticeable

The Bottom Line

Good antibody for using as phenotyping marker or compensation control but probably not for measuring expression kinetics.
