PE or phyco-erythrin is my favorite fluorophore and I used it for detecting CD4 T cells within mouse splenocytes. Thus far, this was the best staining I have ever seen. The fluorescence shift was optimal, no background or bleeding and the populations were relevant. A highly recommended antibody; perhaps a highly recommended fluorophore as I have noticed it to work beautifully with pretty much any target.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse splenocytes

Primary Incubation

25 minutes on ice

Blocking Agent

CD16/32 antibody for blocking Fc receptors

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow cytometry on PE channel

Results Summary

The staining worked well with about 2 log increase in fluorescence of positive cells. The background was minimal and frequencies of positive and negative cells were physiologically relevant.

The Bottom Line

PE is the most desired fluorophore. If you have it available in your panel, then use it for any target.
