We tried several electro competent cells to make clonal library construction for genome analyses. We found CloneCatcher electro competent cells to be good with good transformation efficiency to capture the diversity from the ligation mixture.

Ligation mix
The CloneCatcher electro competent cells showed high efficiency transformants when we electroporated 2-3ul of concentrated ligation mixture in to 25ul of the cells. We followed recommended electroporated conditions as comes with manual. It needs purification of ligation mixture and elute it in water before doing electroporation one key to get successful transformation.
Avoid growing culture more than 16hrs
We made library with lenti viral vector and obtain transformation efficiency and diversity in our library preparation.
