In our lab we perform a lot of molecular biology and protein expressions. The DS-11 spectrophotometer is really great for measuring DNA/RNA/ and protein concentrations. The fact that it has an easy to use interface makes it fun to use. For example, it is easy to switch between DNA and protein concentrations. The DNA concentration has an auto measure capability so all you do is quickly kim wipe the nano drop and place your sample (.5 to 2 ul). The same rules apply for protein concentration. As long as you know what the molecular weight and extinction coefficient is then it can calculate the mg/ml for you. The only draw back is the fact that it does not tell you what path length it uses when measuring protein concentrations which makes it hard to double check the calculations. You just have to trust that it adjusts the path length. Otherwise very useful instrument and highly recommended. -Nancy Hernandez (Sagar Khare Lab at Rutgers University)

Take 2 ul of Elution buffer to blank the instrument. Then another 2 ul of the DNA sample and hit read.

Tips

Try to use 1 ul or 2 ul (easy to handle)

The instrument tells you the ng/ul after taking into consideration the blank from before and even calculate the ratio of A260/280 which tells you how pure the sample is.

Summary

The Good

Easy to use for DNA/RNA/Protein Concentrations. Also the calculator has a Tm measurement along with GC content so it's great for primers.

The Bad

We would have liked to know the path length used during the protein concentration measurements so that we may double check the measurement.

The Bottom Line

Fantastic instrument and highly recommended for any lab that heavily uses molecular biology and protein expression techniques/protocols.
