This antibody was used in a staining panel to identify Tregs in the thymus and the spleen. The CD25-Brilliant Violet 421 antibody had a great noise-to-signal ratio and also withstood the fixation and permeabilization step required for the Foxp3 staining. Highly recommended.

Results Summary

The new Brilliant Violet 421 dye is a much brighter fluorochrome than Pacific Blue or eFluor 450. Pairing this bright fluorochrome with a dim marker like CD25 gave us an excellent separation between CD25- and CD25+ cells. No issues with compensation with other channels. If you are currently using BD Compensation beads to compensate, this antibody will not bind to the rat beads as the light chain is lambda and not kappa. It is best to compensate with thymocytes or try the OneComp beads. This antibody also works wells with fixation and permeabilization buffers needed for Foxp3 staining from eBioscience.
