HBSS was used to extract spleens from mice and create a single cell suspension of splenocytes in HBSS media.

Protocol Overview

Spleens from mice are removed and placed into HBSS containing 1% FBS. Spleens are then crushed through mesh, and single cell suspension is created using a 5ml syringe and 18 gauge needle. The suspension is then centrifuged, red blood cells are lysed, cetrifuged again, and then brought up in HBSS for counting.

Tips

Addition of 1%FBS helps to create a more stable environment for the cells when they come out of the mouse.

Results Summary

The HBSS works as a great medium when harvesting mouse tissue or organs. It gives the user the ability to get the cells into culture in a timely manner without having a great loss of cells.
