Company:

BioLegend

Product Name:

APC anti-mouse TNF-α Antibody

Catalog Number:

506308

This antibody was used for intracellular staining of TNF-alpha on mouse CD8 T cells that were stimulated with OVA peptide. This seems to be a strong stimulation as most cells were positive for TNF-alpha after 3 days of stimulation. Usually, with a polyclonal stimulus, only a fraction of cells may express TNF-alpha. I recommend using different stimulation conditions to standardize the staining of this antibody.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Purified mouse CD8 T cells

Primary Incubation

1:200 dilution for 1 hour on ice

Blocking Agent

NA

Secondary Incubation

NA

Tertiary Incubation

Detection

Flow cytometry on APC channel

Results Summary

The results show uniformly high expression of TNF-alpha on all cells, which is unexpected. There may be some background staining, needs optimizing.
