Our studies concentrate on identifying T cell memory subsets. The initial step in the identification of these cells is staining for CD4 and CD8. We have been using a very good CD4/CD8 colour combination. For CD4, we use the PE/Cy7 anti-human CD4 Antibody from BioLegend, which gives us very good staining and the resultant CD4/CD8 dot plot is very clean!
Human lymphocytes
5ul per 1x10^6 cells for 45 min at 4 C
Human IgG solution (15 min prior to incubation with antibody)

LSR-II Flow cytometer
The results are shown in the graph where CD4 is on the x-axis and CD8 on y-axis. You can see very distinct CD4 cells, CD8 cells and a few double negatives (DN).
