PCR Cloning Kits

PCR cloning kits are available from a number of suppliers for fast and easy cloning of PCR amplicons directly into a vector. PCR cloning omits the need for restriction enzymes, as it can ligate the vector and DNA fragment of interest through a master mix for ligation. Popular PCR cloning methods include TA cloning and blunt-end cloning. TA cloning uses Taq polymerase that adds an extra deoxyadenosine at the 3' end of the amplicons, which can be cloned into the vector containing complementary 3' deoxythymidine overhangs. Blunt-end cloning allows for the ligation of DNA fragments that do not contain overhangs. PCR cloning kits often contain specialized classes of vectors, such as TOPO vectors. The vectors incorporate selection markers, such as genes for antibiotic resistance, and some possess universal sequencing primer sites. Certain kits contain competent cells, while others do not.
