| Application |
| This product is especially tested and qualified for use in all PCR or
RT-PCR reactions, in conjunction with conventional thermocyclers or with
the LightCycler Instrument available from Roche Applied Science. All LightCycler
Kits – available from Roche Applied Sci-ence – and some of the Amplification
Products – available from Roche Applied Science – contain this Water, PCR
Grade. |

| Storage and Stability |
Store a working solution for a short period of time
(e.g. 1 week) at 2 - 8 °C Store stock solutions for a longer period of time

| Quality Control |
| Test buffer: 10 mM Tris/HCl, pH 7.5 (37°C), 10 mM MgCl 2 , 1 mM DTE |
| Absence of endonucleases: |
1 µg
DNA is incubated with various amounts of Water, PCR Grade in 50 µl
test buffer at 65°C for 16 h. Up to 20 µl Water, PCR grade shows no
detectable degradation of the DNA. |
| Absence of endonucleases: |
1 µg DNA,
cleaved by Eco RI and Hind III (MWM III) is incubated with various
amounts of Water, PCR Grade in 50 µl test buffer at 65°C for 16 h.
Up to 20 µl Water, PCR grade shows no detectable degradation of the
DNA fragments. |
| Absence of nicking activity: |
1 µg supercoiled pBR322 DNA is incubated with vari-ous
amounts of Water, PCR Grade in 50 µl test buffer at 65°C for 16 h.
Up to 20 µl Water, PCR grade shows no detectable alteration or relaxation
of the supercoiled DNA. |
| Absence of ribunocleases: |
5 µg of MS2 RNA are incubated with various amouts of
Water, PCR Grade in 50 µl test buffer at 37°C for 1 h. Up to 20 µl
Water, PCR Grade shows no detectable RNAse activity. |
| Amplification function test: |
Each lot of Water, PCR Grade is function tested in
PCR. Routinely, PCR is performed using DNA
for an 0.5 kb fragment. A clear defined band from 0.01 ng DNA
should be clearly visible. |
