DNA Loading Dyes

DNA loading dyes are used during gel electrophoresis for smooth loading of DNA into wells and for tracking DNA migration across the gel. They are used for both agarose and native polyacrylamide gels. The composition of the dye can vary but will usually contain some of the following tracking dyes: Bromophenol Blue, Orange G, or Xylene Cyanol FF. Varying concentrations of DNA loading dyes are available, and the concentration can also be adjusted according to experimental needs. EDTA may be included in some dyes as a chelating agent and to inhibit nuclease activity, while glycerol helps the DNA in the ladder and sample form a layer at the bottom of the well.
