The mouse cell line was treated with a growth factor examined to test its effect on the steady-state level of UCP1 proteins. For this, growth-arrested mouse cells were treated with the growth factor, and cells were lysed in RIPA buffer. The proteins were extracted, estimated, and resolved in 10% SDS-PAGE. Resolved proteins were western transferred and immunoblotted with UCP1 antibody. The selection of the UCP1 antibody from Abcam was based on the colleague’s previous experience with the same antibody.

Primary Incubation

Dilution (1:1000) in 5% skimmed milk prepared in 1x TBST, overnight incubation at 4 degrees celsius.

Blocking Agent

5% skimmed milk made in 1x TBST.

Goat anti-Rabbit IgG HRP Ab, dilution (1:4000) in 5% skimmed milk made in 1x TBST, 1hr at RT.

Tertiary Incubation

None

Detection

Enhanced chemiluminescent detection in X-Ray films.

Results Summary

An excellent, bright, and highly specific band of UCP1 was detected at nearly 36 kDa by Abcam antibody. No non-specific protein bands were observed.
