I use this Fc block for a variety of staining purposes, mainly to stain monocyte-derived dendritic cells for flow cytometry. It has never let me down!
Flow cytometry
Human dendritic cells
0.1uL Zombie Green viability stain in 100uL PBS, incubated at room temp for 15 mins.
Human TruStain Fc block, 5uL diluted in 95uL FACS buffer (5% FBS in DPBS), incubated at 4C for 10 mins
Desired antibody (as needed).
Cytofix buffer, 100uL for 20 mins at 4 degrees C.
Flow cytometry in a BD Celesta cytometer.
This Fc block has helped block Fc receptors for a variety of purposes, including a recent study on human primary dendritic cells in which I was looking for confirmation that cells were activated post-treatment by examining activation markers. In this study, almost all cells (>95%) exhibited activation markers CD80/86/40, with no non-specific binding.
