The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. This experiment aims to determine good working antibodies to blot some of the core components of the WNT pathway. This antibody picks up two bands and is excellent for Western blotting.

Experimental Design and Results Summary

Applications

Western blot

Sample

RKO whole cell extract

Primary Incubation

1:2000 dilution; 16 hr at 4 deg C

Blocking Agent

5% Milk in TBS-T (0.1% Tween-20)

Secondary Incubation

Donkey anti-Rabbit IgG HRP used at 1:10,000 dilution; 1 hr incubation at RT

Tertiary Incubation

None

Detection

ECL regular

Results Summary

Whole cell extracts from RKO (human colorectal cancer cell line) was resolved on 4-12% Bis-Tris gel followed by protein transfer to nitrocellulose membrane. This antibody detects two bands at ~58 and ~79 kDa. TCF7L2 null line served as a good control in this experiment.
