ACK (Ammonium-Chloride-Potassium) Lysing Buffer is used for the lysis of red blood cells in samples containing white blood cells, such as EDTA-treated whole blood, buffy coats, and bone marrow. Red blood cells are permeable to ammonium chloride, which causes cellular swelling and eventual rupture of the cell membrane. I usually use it when I need to remove RBC to isolate either buffy coat or platelets.

Pipette 1mL EDTA-treated whole blood into a tube containing 10 mL of Gibco™ ACK Lysing Buffer at room temperature and invert the tube a few times. Allow the blood sample plus ACK Lysing Buffer to incubate at room temperature for 3 – 5 minutes. Add an equal amount of PBS to block the reaction and wash and then centrifuge. Lysis of the red cells should be evident during this incubation. If you still have a lot of red blood cells repeat the passage describe above and eventually reduce the volume of ACK so you don't harm the cells.
