Cells at 80% confluency in 35-mm cell culture dishes are lysed in ice-cold 100 μl RIPA buffer containing 1 μl protease inhibitor cocktail. Supernatant from cell lysates is used for gel electrophoresis followed by western blot. Nitrocellulose or PVDF membrane containing protein bands are blocked and incubated with primary and secondary antibodies. The protein bands are then detected using SuperSignal West Pico Chemiluminescent Substrate in ChemiDoc MP Imaging System.
