We generated a knockout of a member of the JAK family of proteins and wanted to examine whether there were changes in EGFR signaling. We examined ophospho-Y1068 EGFR signal in control and JAK knockout (KO) cell lines.

1:1000 dilution of primary antibody in 1% BSA incubated overnight at 4 degrees celsius on a slow rocker.

1:1000 goat-anti-rabbit HRP conjugated antibody suspended in 1% milk for one hour at room temperature on a slow rocker.

We found that phospho-Y1068 EGFR signalling was only elevated in one JAK KO cell line. Because this amplification was inconsistent, we concluded that this was likely a clonal cell line difference and not attributable to the knockout of the JAK protein.
Robust bands in expected size range.
If anything, the signal is so robust that it can become overexposed when using film.
This is an excellent antibody for phospho-Y1068 EGFR!
