FITC Annexin V binds to phosphatidylserine and enables the identification of procoagulant platelets, which express phosphatidylserine and participate in coagulation. Annexin V staining can be challenging and requires the addition of calcium to staining buffers.

Results Summary

Citrate-anticoagulated human whole blood was diluted 1:10 in HEPES-Tyrodes-BSA containing (1) 2.5 mM calcium to facilitate FITC Annexin V binding, (2) 2.5 mM Gly-Pro-Arg-Pro (GPRP) to inhibit clot formation, (3) 1 µg/mL FITC Annexin V, and (4) 10 µg/mL CRP-XL and 10 µM TRAP (platelet agonists) to induce platelet activation and phosphatidylserine exposure. Lyse/fix buffer was added, and samples were acquired by flow cytometry. FITC Annexin V allowed clear resolution of procoagulant platelets.
